I defrag about once a month on average. I don't use disk cleanup, though. I use C-Cleaner for disk cleanup. I find it works a LOT better.
You can get it here: http://www.ccleaner.com/download
I run it every 3-4 days. Try it out. It's nice and easy too. Just click Run Cleaner and let it do its thing.

I would just use it every few days then. I use it and don't bother with disc cleanup. I find it is more effective, faster and a reliable way to help keep things running well.

Disk defrag? Not going to affect a browser's operation. Ccleaner will clean temp files which may help. What browser are you using? If it's internet explorer download firefox and try that. You could also try to repair IE through control panel > windows programs ect..

System restore to when it was working, if it's a conflicting 3rd party progrma or an update this should take care of it, if not open IE7 and go to tools > internet options > advanced then click the reset button, it'll dump any passwords and stuff that you have saved though so be sure you know them before you click the button
